I'm sure glad you chimed in on this subject Al........I was starting to think I was the only one here that even cared or believed in the Amyloid Cascade and this study just goes to help, once again, prove the Amyloid-beta Theory and everything I've been posting here relentlessly. There is an overwhelming body of evidence in the order of hundreds of Studies showing the order in which the events occur.
One that comes to mind is Joanne Mclaurin's Study (2000)......
Dr. Cruz Quote explains it in a nutshell........."Dr. Joanne Mclaurin (Discoveror AZD-103) was looking at Neuronal Toxicity where Amyloid binds to Neuronal Cells and kills the Neuronal Cells and what she found was that if she added AZD-103 it PREVENTED the Neuronal Cell Death. Then what she found was that to create Neuronal Cell death you need Amyloid Fibrils (Oligomers) to come into contact with the Neurons. Just free Amyloid (Monomers) is not sufficient. What she found was that when she added AZD-103 was that it Broke Up the Fibrils."
TAU is a DownStream Event in the Amyloid Cascade. AFTER Inflammation and Glial Cell Activation and the resulting OverProduction of the ProInflammatory Cytokines Interleukin-1β ( Il-1b) and Tumor Necrosis Factor (TNF)........this Cytokine Overproduction results in Neuronal Failure and ultimately Neronal Cell Death.

There a large body of evidence showing (and growing) that the memory starts to go even before the Plaques appear (MCI). Very very early Alzheimers. Small Soluble Oligomers (Dimers & Trimers) of Amyloid start to short out the Synapses and the Dendritic Spines lose their shape and become smaller and/or disappear (Spine Loss) just like the study reffered to that you posted where they saw nearby nerve cells changing shape. This imaging Study completely backs it all up!!
Funny thing though is that Transition has TWO of the best candidates to ameliorate these effects. Yup.
1)AZD-103/ELND005 for the Amyloid-b Plaques.
2) MINOZAC to effectively suppress the resulting Over-Production of the Pro-Inflammatory Cytokines IL-1 & TNF that interupts the Synapses electical signals and ends up with the death of the Neurons.
