I had a discussion one day with a leading R&D about that same topic, 7 years ago before he passed told me at that time the cost was 1 billion to take that drug from R&D to the drug store after all is said and done, He could tell me that fact because he did it twice and now is in the hall of fame.  The only shame is he had already passed when they gave him that award and although a humble man I'm sure it would have made him happy.
