RE:Understanding the Absolute benefit at the Interim Data point For me "The DSMB / FDA reset the required patient level to 650 patients" does not indicate an 11% absolute benefit had been achieved. It indicates their hope at the time.
Of course, their numbers are very problematic one way or the other you look at them. But in my opinion, it is harder to believe now that they had reached (although not in a statistically significant way) an 11% absolute benefit with a population on average less sick and finally got a 5% AB with a sicker population, if we are to also believe what they said multiple times that their product was most efficient on sicker patients.
