Time to Market for H2S Pipeline

That's all that matters.

P2b/3a for OA to prove safety and efficacy for chronic use will be time and resource intensive.

P2b/3a for Post-Operative Pain for 10-14-day course of Otenaproxesul at 150 mg PO daily is ready to go with supporting AME data.  Post-op mild LTE vs. GI ulcers, no one cares about 3-5x LTE in small subset especially when self-limiting because bigger concern is post-op DVT prophylaxis (particularly orthopedic surgeries) where incremental risk of GI bleeding with NSAIDs + DVT prophylaxis is a far worse outcome, and stopping prophylaxis due to bleeding is even worse.

Fast-track Otenaproxesul as a GI-safe opioid-sparring agent for mild-to-moderate post-operative pain as a preventative strategy for opioid addiction.

Once it's on the market, all subsequent INDs/CTAs will go faster.  Overall this is a much better drug developement strategy imho.  GLTA.

You're making a huge assumption re H2S's effect on the liver. The supposed toxicity you;'re alluding to is elevated LTEs, which is a class issue with NSAIDs and indeed ALL other medications on a longer time horizon. Protect and yet we got thrown out of AME for having high LTE. The rate is higher than regular nsaids. If it is protective as what you are claiming we would have lower LTEs than other nsaids. Don'y give false hope to people

My take is it's on par with other NSAIDS.  In fact from an API perspective, it's less NSAID being ingested. Why are LTE's occurring then?  H2S may be inhibiting cytochrome p450 enzymes responsible for NSAID metabolism therefore reduced clearance of metabolites through kidneys in long-term use.   Since dose-dependence has been confirmed, the dose needs to be ...more

Mstrmind, I'll look it up but there is parti cyp 450 inhibition. I mentioned it in ceo but I believe it is the c9 pathway (don't quote me til I find the study). It almost certainly is building up due to his inhibition, but can also almost certainly be corrected by dropping the dose and requiring a longer ramp up time before we see clinical significance. Yes, completely agree.  This was briefly discussed at the AGM as well by Stauffer although at that time awaiting biomarker confirmation.  150 mg x 90 days will not work due to p450 inhibition.  Loading could work although challenging for OTC marketing.   150 mg x 14 days works despite p450 inhibition, both safe and effective, hence new strategy. I doubt they would go with  dosing regimen (Q8H) as part of their differentiation strategy was being "a once a day pill".  Ill say that their research into selecting a different moiety besides TBZ will be a major factor in keeping that OD differentiation.

H2S is not toxic, review the 2a and 2b datasets.  5/42 people with LTE when 3/42 with LTE were expected on a dose ranging AME study does not equal toxicity.  There are numerous variables at play and external factors that cannot be ruled out.  The business strategy is clear, data to support short-term or PRN use is there, why go long-term use for OA given AME findings?  That ...more

We got withdrawn but the company said it nicely. If we have same lte like other naaids. The AME would be resumed. I got out with 80% loss and wont be a sheep and believe what dan rights

No one is spreading false information but you guys. Don't  give false hope to people. Atb 346 is out and the AME is not published because it is shi*t. They only mentioned about the 5x which triggered the stop and i gairantee you everyone has elevated LTE (x2, x3,x4).  If only a couple of people has elevated lte they would not move to acute pain which is 2 weeks treatment. the stomach ...more

This is what the company has reported in AGM and recent release.  There's no need to speculate anything otherwise, especially negative.  The market has not responded favourably, but make no mistake Antibe has a Phase 3 drug for post-operative pain coming and osteoarthritis is still on the table, with IBD next.
