PDC's

Currently there are only 2 PDC's that have FDA approval (article is one year old).

In my view, Melflufen in not an PDC. It's a prodrug. It's not aiming at a cellular target, the added amino acid is there just to increase the lipophilicity and allow the prodrug to enter the cells. Once inside the cells, the prodrug is cleaved in two parts releasing the free drug. It has nothing of the targeted element and selective distribution associated with the principle behingd PDCs ...more

By the way, it seems Melflufen has beed withdrawn from the market because of excess deaths in phase III confirmatory trial. So it is now irrelevant to argue if it's an approved PDC or not, it is no longer approved. Always remember that in cancer you can have fast approval, but confirmatory trials are requested for long term safety and to gather data on a higher number of patients. https://www ...more
