AGEs are slowly formed by nonenzymatic condensation of reducing sugars with amine residues on proteins, lipids, or nucleic acids. Tissue AGEs increase with chronologic age, oxidative stress, inflammation and diabetes. AGE deposition in tissues is thought to contribute to the development of diabetes complications. The objective was to compare skin AGEs from new onset T2D patients with obese (BMI z-score > 1.45 SD) children without diabetes (HbA1c<6) from clinic. Skin AGEs were estimated by measurement of skin intrinsic fluorescence (SIF) using a SCOUT DS (VeraLight, NM) at excitation wavelengths of 405 and 420nm. Raw fluorescence was corrected using excitation (kx=0.5) and emission (km=0.5) constants previously found not influenced by race. SIF is in arbitrary units (AU). SIF was measured within 1 mon of T2D diagnosis. SIFs (dependent variable) were analyzed in separate multiple variable models with group (diabetes or obese), chronological age, BMI (z-score) and gender as independent covariates.
